You can upgrade your IBM xSeries 370 8681-3RX upto a maximum of 16/32 GB, with maximum of per slot in 16 (4 banks of 4) memory slots. The IBM xSeries 370 8681-3RX takes the PC100 Registered ECC SDRAM DIMMs. To support over 16 GB, the optional memory card must be installed. It must be configured identical to the standard memory card to achieve memory interleave. Memory Type.

So you must add memory four pieces at a time (and they must be the same size and type of memory) in order for the system to benefit from the upgrade.
